Antibody medicines for obesity & cardiometabolic disease

Today’s therapies press the accelerator. We’re building medicines to release the brake.

Engineering precision obesity medicines for better, healthier, more sustainable weight loss.

Our approach

Body weight has two opposing systems

GLP-1 medicines have transformed obesity care by working the accelerator. Overture is focused on the other side of the equation.

Accelerator pedal
The accelerator

Nutrient-stimulated hormones increase satiety, helping people eat less and lose weight.

Brake pedal
The brake

Evolutionary biology protects fat stores by decreasing satiety and slowing energy expenditure.

Single gene one inactive copy
Human genetics reveals the brake

Some people are born with less brake

People who naturally carry an inactive copy of a target gene are leaner and metabolically healthier. Overture aims to learn from and replicate this natural protection.

  1. 01

    Releases the Brake

Binds the native receptor and locks it in an inactive state, blocking downstream signaling.

  2. 02

    Long-Acting Potential

Fc-engineered for half-life extension, targeting monthly-to-quarterly subcutaneous dosing.

  3. 03

    Highly Specific

Low-nanomolar binding with no off-target hits across cell surface membrane proteome.

  4. 04

    Complementary

Acts on orthogonal biology that can add to nutrient-stimulated hormones rather than compete with them.

Pipeline

A foundation for multimodal care

Obesity is biologically diverse, and many patients will be best served by more than one mechanism. OVT-109 is built as a core antibody that can stand alone, pair with incretins, or anchor long-acting multi-specifics.

Illustrative profiles across eight metabolic parameters Octagon chart comparing a GLP-1 receptor agonist, OVT-109, and the OVT-109 plus incretin combination across satiety regulation, anti-steatosis, anti-inflammatory effect, insulin sensitivity, visceral fat loss, triglyceride and LDL lowering, energy expenditure and skeletal muscle retention.
